How can I enable different region otherthan East US and North Europe? And also do we need to have atleast 1 server on standby for matchmaking with server allocation?

Please check this documentation - Managing quota changes - PlayFab | Microsoft Docs to learn about how to request the quota changes. Quota changes can include 1 or more changes to a VM family and region combination.

Yes, if you need to test matchmaking with server allocation, you need to have at least one server build and one standby server instance. You can check the documentation - Integrating with PlayFab Multiplayer Servers - PlayFab | Microsoft Docs how to integrate the matchmaking with PlayFab Multiplayer Servers.
